Best Life Gym, off Marlow Road, was unveiled by the Mayor Lorraine Hillier and her sister and Mayoress Susan George on Saturday.
Almost 30 members of Hourglass Performance Training, a women's fitness club based in Stoke-on-Trent, were first to try the equipment.
They had read about the gym's launch on the internet and asked if they could attend as they were fans of Best.
Before starting their workout, the women queued up to take “selfies” with him. He then showed them how to use the equipment and gave them some exercise tips.
Best, 34, is the son of the late footballer George Best and personal trainer Angie Best, who owns the neighbouring Metaphysical gym at Henley Business School. Mrs Best, 63, who lives at Bix Common, also attended the launch.
Best, who grew up in Los Angeles and now lives in London, came up with the idea after visiting his mother’s gym and decided to go ahead when the LA Fitness gym in Newtown Road, Henley, shut in August.
His premises, which belong to the business school, used to be a squash court. They now have free weights, resistance machines, exercise bikes and various boxing pads and bags.
Best will be there three days a week while Mrs Best will attend daily.
He said: "I considered opening a gym in London but I'd been in London for about 15 years and Henley is such a beautiful place.
"The air feels so much cleaner and fresher out here and the people are lovely so it seemed like the ideal venue.
"I'm very proud of what I've created and couldn't be any happier. We've had a lot of interest from all over the community.
“I want to expand but I'm very proud that my flagship gym is in Henley."
Councillor Hillier said: "It's an amazing space with a lot of equipment. I hope lots of people will use it. My sister and I certainly intend to, even if we're not dressed for it today!"
